Medeama head coach Yaw Preko took charge of his game since replacing Samuel Boadu who left for Hearts of Oak after the first round of the league.
Black Satellites player Samuel Ashie Quaye scored the only goal of the game in the 41st minute.
Preko – who also briefly coached Olympics this season – handed starting places to new signings Ivorian full-back Oumar Coulibally, Zakaria Mumuni and former Hearts of Oak defender Vincent Atingah as Prince Opoku Agyemang and Ahmed Toure started up front.
Opoku Agyemang should have done better after a Toure cross found him in the Olympics area but the striker who had the best option of squaring the ball for Mumuni fired a low shot that was easily stopped by goalkeeper Benjamin Asare.
Medeama are thus winless against Olympics this season as they drew at home to their Friday opponents in Tarkwa before this latest defeat.
